Studio Usa: attraverso il litio è possibile bloccare la proteina Gsk 3 responsabile dell’invecchiamento e attivare la molecola Nrf2 che difende contro i danni alle cellule


Per ora gli esperimenti sono stati condotti sugli insetti, ma sembra che promettano l’arrivo di un potente elisir di lunga vita: una pillola anti-invecchiamento che potrebbe aiutare a vivere fino a una decina di anni in più. Si tratta di un grande avanzamento nello studio, che va avanti da alcuni anni, di una proteina chiamata Gsk-3 (glicogeno sintasi chinasi 3).

Essa è in grado di accorciare la vita naturale di un essere vivente, ma può essere inibita attraverso una sostanza nota per regolare l’umore in chi soffre di disturbo bipolare: il litio.

Ebbene, negli studi condotti dagli scienziati dell’Ucl Institute of Healthy Ageing (Usa), del Max Planck Institute for Biology of Ageing e dell’European Molecular Biology Laboratory sui moscerini della frutta - che possiedono anche loro la proteina Gsk-3 - si è dimostrato che somministrando basse dosi di litio, si riesce a estendere la vita degli insetti in media del 16%.

Questo ha sollevato speranze che si possa arrivare a una pillola a base di questa sostanza - o più probabilmente un altro farmaco simile, con meno effetti collaterali - in grado di prolungare l’esistenza umana. E, cosa forse più importante, «la conoscenza di questo meccanismo potrebbe anche rivelare il segreto per evitare l’insorgenza di malattie legate all’età: Alzheimer, diabete, cancro e Parkinson», ipotizza Jorge Ivan Castillo-Quan, autore principale dello studio che appare su Cell Reports.

Il team ha rilevato in particolare che il litio blocca l’invecchiamento bloccando Gsk-3 e attivando un’altra molecola chiamata Nrf2, che si trova nei vermi, nelle mosche e nei mammiferi (compreso l’uomo), e rappresenta un’importante difesa contro i danni delle cellule.

L’esperto avverte comunque che una pillola che bersagli Gsk-3 è improbabile che venga resa disponibile entro i prossimi 10 anni, anzi: potrebbe non arrivare prima di diversi decenni. Ma una volta messa a punto, potrebbe potenzialmente aumentare la durata della vita di circa 7-10 anni. I ricercatori ora vogliono condurre studi su animali più complessi come i topi, prima di passare ai primati e poi agli esseri umani.
