I tardigradi possono resistere per anni alla completa disidratazione grazie a particolari proteine che impediscono la cristallizzazione del contenuto delle cellule, favorendo invece la formazione di una struttura vetrosa. La scoperta può avere diverse applicazioni, come la conservazione senza la catena del  freddo di farmaci sensibili al calore.

E' un particolare gruppo di proteine che permette ai tardigradi di sopravvivere anche dieci anni a uno stato di disidratazione estrema. A scoprirlo è stato un gruppo di ricercatori dell'Università del North Carolina a Chapel Hill, in collaborazione con ricercatori dell'Università della California a Berkeley e dell'Università di Modena e Reggio Emilia, che firmano un articolo pubblicato su "Molecular Cell".

I tardigradi - minuscoli invertebrati che prediligono gli ambienti acquatici - hanno la capacità di sopravvivere in  ambienti estremi che distruggerebbero la maggior parte degli altri esseri viventi. Sono infatti in grado di resistere per anni al congelamento, all'esposizione al vuoto e alle radiazioni dello spazio e all'essiccazione.

Studi su altri organismi che sono in grado di resistere a gravi stati di disidratazione - per esempio i lieviti, alcuni nematodi e le uova del crostaceo Artemia salina - avevano suggerito che anche la capacità  di sopravvivere alla mancanza di acqua dei tardigradi fosse legata a uno zucchero, il trealosio. Successive analisi hanno però mostrato che i livelli di questo zucchero nei tardigradi sono estremamente bassi se non nulli.

Thomas C. Boothby e colleghi hanno quindi deciso di monitorare i livelli di espressione dei geni di questi animaletti in diverse condizioni: allo stato naturale, quando venivano congelati e quando venivano essiccati. I ricercatori hanno scoperto che i geni più sovraespressi durante l'essiccazione, sono quelli che codificano un particolare gruppo di proteine, dette TDP (tardigrade-specific intrinsically disordered proteins) che fa parte della classe delle cosiddette proteine intrinsecamente disordinate che, diversamente dalle altre proteine, non hanno una struttura 
tridimensionale fissa.

Il meccanismo grazie al quale le TDF permettono la sopravvivenza dei tardigradi all'essiccazione è simile a quello del trealosio, ma ancora più efficiente: quando un organismo perde completamente l'acqua che contiene, le TFG impediscono che le molecole presenti all'interno delle cellule si aggreghino formando una struttura cristallina (che distrugge le caratteristiche strutturali di cellule e tessuti), e favoriscono - grazie proprio alla loro struttura tridimensionale non definita - la formazione di solidi "vetrosi" che conservano la struttura cellulare originaria.

Secondo Boothby, le TDP hanno un elevato numero di potenziali applicazioni, dalla protezione delle colture dalla siccità, alla conservazione di farmaci che normalmente richiedono uno stoccaggio a freddo.  "Sono cresciuto in Africa, dove in molte aree remote la mancanza di refrigerazione è un problema enorme. La possibilità di questo  tipo di applicazione è una delle cose che mi hanno spinto a studiare i tardigradi", ha detto Boothby.

Thomas C. Boothby and colleagues therefore decided to monitor the levels of expression of the genes of these animals in different conditions: the natural state, when they were frozen and when they were dried. The researchers found that the genes most overexpressed during drying are those which encode a particular group of proteins, called TDP (tardigrade-specific intrinsically disordered proteins) that is part of the class of so-called intrinsically disordered proteins that, unlike other proteins , have no structure
