The relationship between immune system and cancer is elaborated and dynamic.
Immunotherapy has been acquiring big visibility in recent years and even more in the last months thanks to the 2018 Nobel Prize for medicine. Immunotherapy involves re-engineering T-cells so that they can recognize and kill cancer cells. This type of therapy has shown promising preliminary results in fighting lymphoma.
Microscopy is the elected method to study immune cells interactions and their potential for cancer healing. However, technologies that are available nowadays mainly rely on fluorescence, and fragile primary immune cells are heavily perturbed and eventually killed by staining procedures and phototoxicity.
The research on immune-oncology will greatly benefit from a high-resolution, label-free approach allowing to image immune cells interplays in a marker-free, non-invasive way.
Nanolive’s 3D Cell Explorer opens the door for new observations and for long-term live cell imaging (up to weeks of continuous imaging) with unprecedented spatio-temporal resolution (<200nm; 1img/2sec).

APC T-cell interaction/activation experiment
In the first movie obtained with Nanolive’s 3D Cell Explorer, we can witness to the T-Cell activation process totally marker-free. Pre-stimulated antigen presenting cells (APCs*: dendritic cells and macrophages) are cultured with freshly isolated “naïve T-cells”, the OT-I mice T-cells in order to teach them to recognize and kill tumor cells.
* APCs are special cells that present the antigens on their surface so that other immune cells can “see” and detect it.



Video 2: Activated T-cells were incubated against the OVA antigen were incubated with MC38-OVA cancer cells expressing the OVA antigen. T-cells killing cancer cells
In this movie and in the zoom-in below you can see T-Cells killing cancer cells.
The cancer cell line is MC38-OVA, a transduced colon cancer cell line that expresses the ovalbumin (OVA) model antigen.
T-cells, coming from OT-I mice, carry a transgenic T-cell receptor responsive to OVA residues 257-264 (SIINFEKL peptide) in the context of the MHC I H2kb.
In this experiment, the T-cells that were activated in the first experiment and that are now called “effectors”, are incubated with MC38-OVA cancer cells. Upon recognition of their target (the OVA residues on the MHC I H2kB of the cancer cells), T-cells induce the killing of the cancer cells. However, this does not happen all the time and many cancer cells survive.

La relazione tra sistema immunitario e cancro è elaborata e dinamica.
L'immunoterapia ha acquisito grande visibilità negli ultimi anni e ancor più negli ultimi mesi grazie al Premio Nobel per la medicina 2018. L'immunoterapia comporta la reingegnerizzazione delle cellule T in modo che possano riconoscere e uccidere le cellule tumorali. Questo tipo di terapia ha mostrato risultati preliminari promettenti nella lotta contro il linfoma.
La microscopia è il metodo eletto per studiare le interazioni delle cellule immunitarie e il loro potenziale per la guarigione del cancro. Tuttavia, le tecnologie disponibili oggigiorno si basano principalmente sulla fluorescenza e le fragili cellule immunitarie primarie sono fortemente perturbate e infine uccise dalle procedure di colorazione e dalla fototossicità.
La ricerca sull'immunologia oncologica trarrà grande beneficio da un approccio innovativo e ad alta risoluzione che consente di acquisire immagini delle cellule immunitarie in modo non distruttivo e non invasivo.
3D Cell Explorer di Nanolive apre le porte a nuove osservazioni e all'imaging cellulare a lungo termine (fino a settimane di imaging continuo) con una risoluzione spazio-temporale senza precedenti (<200nm; 1img / 2sec).
Esperimento di interazione / attivazione di cellule T APC
Nel primo film ottenuto con 3D Cell Explorer di Nanolive, possiamo assistere al processo di attivazione T-Cell totalmente privo di marker. Le cellule presentanti l'antigene pre-stimolato (APC *: cellule dendritiche e macrofagi) sono coltivate con cellule T naïve appena isolate, le cellule T dei topi OT-I per insegnare loro a riconoscere e uccidere le cellule tumorali.
* Le APC sono cellule speciali che presentano gli antigeni sulla loro superficie in modo che altre cellule immunitarie possano "vederle" e rilevarle.
Le cellule T uccidono le cellule tumorali

La linea cellulare cancerosa è MC38-OVA, una linea cellulare trasdotta del cancro del colon che esprime l'antigene del modello dell'ovaalbumina (OVA).Le cellule T, provenienti da topi OT-I, trasportano un recettore di cellule T transgenico sensibile ai residui di OVA 257-264 (peptide SIINFEKL) nel contesto del MHC I H2kb.
In questo esperimento, le cellule T che sono state attivate nel primo esperimento e che ora vengono chiamate "effettori", sono incubate con cellule tumorali MC38-OVA. Dopo aver riconosciuto il loro obiettivo (i residui di OVA sul MHC I H2kB delle cellule tumorali), i linfociti T inducono l'uccisione delle cellule tumorali. Tuttavia, questo non accade sempre e molte cellule tumorali sopravvivono.
