La scoperta di un sistema batterico di difesa dai virus ha portato a una nuova tecnica di editing simile a CRISPR ma che interviene sull'RNA invece che sul DNA. A differenza di CRISPR, che produce cambiamenti permanenti nel genoma, la nuova tecnica chiamata C2c2 produce modifiche temporanee.

Un sistema di editing simile a CRISPR, ma che ha come bersaglio l'RNA al posto del DNA, è descritto in un articolo su "Science". A differenza dell'intervento sul DNA con CRISPR, che produce cambiamenti permanenti nel genoma, l'intervento sull'RNA con il nuovo sistema potrebbe portare a modifiche temporanee. Inoltre, il nuovo approccio, indicato con la sigla C2c2 (da  Class 2 type VI-A CRISPR-Cas effector), agisce con maggiore specificità e funzionalità rispetto ai metodi attuali come l'interferenza dell'RNA (iRNA), che come dice il nome possono interferire con l'espressione dei geni.

Il nuovo sistema è stato caratterizzato da ricercatori del Broad Institute del Massachusetts Institute of Technology e della Harvard University, dei National Institutes of Health, della Rutgers University a New Brunswick e dell'Istituto Skolkovo per la scienza e la tecnologia, in Russia.

Circa la metà di tutte le specie di batteri usano un sistema di difesa dai virus denominato CRISPR-Cas (Clustered Regularly Interspaced Short Palindromic Repeats and CRISPR-associated genes ) da cui è stato derivato in anni recenti un metodo per modificare il DNA, chiamato per brevità CRISPR. Alcuni virus che aggrediscono i batteri non hanno però un genoma a DNA, invece hanno un genoma a RNA. E ora Feng Zhang, del Broad Institute, e colleghi hanno individuato in un batterio della specie Leptotrichia shahii un meccanismo di difesa da questo tipo di virus simile a CRISPR.

CRISPR agisce su punti specifici della doppia elica di DNA, allo stesso modo C2c2 si dirige specificamente verso un punto del filamento di RNA che identifica con precisione grazie a un "modulo" che porta con sé. I ricercatori sono riusciti sostituire questo modulo così 
da dirigerlo verso specifici RNA di E. coli. Grazie a questa sostituzione, gli scienziati hanno dimostrato che C2c2 può essere programmato per puntare particolari sequenze di RNA delle cellule in modo da inibirne o potenziarne la traduzione in proteine.

Questi risultati suggeriscono una vasta gamma di applicazioni biotecnologiche. Per esempio, la capacità di C2c2 di legarsi a specifiche sequenze di RNA potrebbe essere usata per mettere a punto strumenti di screening su larga scala, creare circuiti cellulari sintetici per la regolazione della produzione di proteine, o anche per marcare con proteine fluorescenti l'RNA stesso e seguirne da vicino l'attività in vivo. "In effetti - ha detto Omar Abudayyeh, primo autore dell'articolo - l'impatto maggiore di C2c2 può riguardare la nostra comprensione del ruolo dell'RNA nel funzionamento cellulare e nell'insorgenza e nel decorso delle malattie."

The discovery of a bacterial defense system against viruses has led to a new editing technique similar to CRISPR but intervening RNA rather than DNA. Unlike CRISPR, which produces permanent changes in the genome, the new technique called C2c2 produces temporary changes.

An editing system similar to CRISPR but that targets RNA instead of DNA, is described in an article in "Science". Unlike intervention on with CRISPR DNA, which produces permanent changes in the genome, the intervention RNA with the new system could lead to temporary changes. In addition, the new approach, indicated by the abbreviation C2c2 (from Class 2 type VI-A CRISPR-Cas effector), acts with greater specificity and functionality than current methods such as RNA interference (RNAi), which tells how the name may interfere with the expression of genes.

The new system has been characterized by researchers at the Broad Institute of the Massachusetts Institute of Technology and Harvard University, the National Institutes of Health, of Rutgers University in New Brunswick and Skolkovo Institute of Science and Technology in Russia.

About half of all species of bacteria use a defense system against viruses called CRISPR-Cas (Clustered Regularly interspaced Short palindromic Repeats and CRISPR-associated genes) from which it was derived in recent years a method to modify the DNA, called for CRISPR brevity. Some viruses that attack bacteria but does not have a genome DNA instead have an RNA genome. CRISPR acts on specific points of the double helix of DNA, in the same way C2c2 is directed specifically towards an RNA strand of the point that identifies with precision thanks to a "module" that brings with it. The researchers were able to replace this form so
to direct it to specific RNA of E. coli. Thanks to this replacement, the scientists have shown that C2c2 can be programmed to point to specific RNA sequences of the cells so as to inhibit or enhance the translation into proteins.

These results suggest a wide range of biotechnological applications. For example, the C2c2 ability to bind to specific RNA sequences could be used to develop large-scale screening tools, create synthetic cell circuits to regulate the production of proteins, or even for marking with fluorescent proteins RNA same and to follow closely the in vivo activity. "In fact - said Omar Abudayyeh, first author of the article - the biggest impact of C2c2 can relate to our understanding of the role of RNA in the cell operation and in the onset and course of disease."
