Alcuni ricercatori hanno prodotto una linea cellulare che rende molto più efficiente la produzione di globuli rossi, e quindi di sangue artificiale. Ecco come.
Con l’aumento della domanda mondiale di sangue per le trasfusioni, sono cresciuti anche l’interesse e lo sforzo per produrre globuli rossiin vitro. Fino a oggi i metodi messi a punto per creare sangue artificiale però erano in grado di produrre solo pochi globuli rossi maturi e richiedevano comunque donazioni ripetute. Oggi invece arriva la notizia che un gruppo di scienziati dell’Università di Bristol e dell’NHS Blood and Transplant è riuscito a produrre globuli rossi su larga scala e più efficientemente di quanto fosse mai stato possibile in precedenza.
“È un grande traguardo – ha dichiarato David Anstee, uno dei principali autori della ricerca – il metodo potrebbe portare, se testato con successo in studi clinici, a una fonte sicura di sangue artificiale utilizzabile per le trasfusioni per persone con gruppi sanguigni rari o in aree del mondo dove le forniture di sangue sono inadeguate o non sicure”.
Per produrre sangue artificiale è necessario partire da cellule staminali in grado di differenziarsi lungo la linea eritroide, quella cioè da cui derivano gli eritrociti (globuli rossi). Il processo di differenziazione non solo è abbastanza lungo e complesso ma, alla fine, produce un numero limitato di globuli rossi. Da qui nasce l’idea dei ricercatori di Bristol, pubblicata su Nature Communication: generare una linea cellulare permanente, che rimanga cioè stabilmente differenziata in cellule eritroidi, ribattezzata Bristol Erythroid Line Adult (BEL-A). Una riserva sempre disponibile da cui ottenere le cellule di interesse.
Le cellule BEL-A riescono infatti a effettuare correttamente tutta l’eritropoiesi, il processo che porta alla produzione dei globuli rossi. In particolare sono in grado di generare reticolociti maturi, ossia elementi di transizione tra gli eritroblasti nucleati e i globuli rossi, corpuscoli normalmente presenti sia nel midollo osseo che nel sangue periferico. Test preliminari hanno anche dimostrato che i globuli rossi prodotti dalle BEL-A hanno un tasso di sopravvivenza simile a quello dei globuli rossi provenienti da donatori.
Come sottolineano gli stessi ricercatori, i risultati fanno ben sperare anche per il trattamento dei malati di talassemia, che possono aver bisogno di trasfusioni ricorrenti di sangue. Nonostante gli incoraggianti risultati comunque, almeno per ora, le donazioni di sangue continueranno a essere fondamentali in tutti gli altri paesi, concludono gli esperti.

To produce artificial blood is necessary from stem cells capable of differentiating along the erythroid lineage, namely that from which they derive the erythrocytes (red blood cells). The process of differentiation is not only quite long and complex, but in the end, produces a limited number of red blood cells. Hence the idea of ​​the researchers of Bristol, published in Nature Communications: create a permanent cell line, that is, to remain permanently in differentiated erythroid cells, renamed Bristol Erythroid Adult Line (BEL-A). A reservation always available from which to obtain the cells of interest.

The cells BEL-A in fact able to carry out properly the entire erythropoiesis, the process that leads to the production of red blood cells. In particular they are able to generate mature reticulocytes, namely transition elements between the nucleated red blood cells and erythroblasts, corpuscles normally present in bone marrow and in peripheral blood. Preliminary tests have also shown that red blood cells produced by BEL-A have a survival rate similar to that of red blood cells from donors.

As the researchers themselves point out, the results also bode well for the treatment of thalassemia patients, who may need to recurrent blood transfusions. Despite the encouraging results, however, at least for now, the blood donation campaign will continue to be fundamental in all other countries, the experts conclude.
