A study coordinated by the University of Milan, in collaboration with the Institute for Research in Biomedicine in Bellinzona, has discovered that ellagic acid, a natural substance found in many fruits and vegetables, limits the formation of biofilms, a protective layer that protects microorganisms from adverse conditions, opening up research opportunities for antibiotic-resistant infections. The publication is in Pharmaceutics.
Ellagic acid, a natural polyphenol found in fruits and vegetables such as walnuts, berries, raspberries, grapes, pomegranates, pistachios, and cashews, is capable of inhibiting the formation of biofilms, a protective layer that allows microorganisms, including pathogens, to survive in less than optimal conditions.
The work, funded by the GSA-IDEA project (University Grand Challenges Project), was recently published in Pharmaceutics and involved a team of researchers from the University of Milan coordinated by Giovanni Grazioso, professor of Medicinal Chemistry at the Department of Pharmaceutical Sciences, and Fabio Forlani of the Department of Food, Nutrition, and Environmental Sciences, in collaboration with Swiss researchers from the Institute for Research in Biomedicine, Università della Svizzera Italiana (Bellinzona).
"Biofilm is a physiological state where multicellular microbial communities are embedded in an extracellular polymeric matrix, allowing microorganisms, including pathogens, to tolerate stressful conditions such as nutrient scarcity, oxidative stress, host defenses, or the presence of antimicrobials such as antibiotics. Precisely because of their resilience, pathogenic biofilms are an important factor in the persistence of both fungal infections, such as candidiasis, and bacterial infections, such as typhoid fever and bloody diarrhea," the team scientists explain.
"The literature does not clarify the mechanism by which ellagic acid acts as an antibiofilm agent, but through computational studies and microbiological, biophysical, and biochemical experimental tests, we have demonstrated that the polyphenol's microbiological activity depends on its interaction with WrbA, an enzymatic protein involved in bacterial biofilm formation. The study revealed that ellagic acid alters WrbA-mediated redox homeostasis by inhibiting its enzymatic function," the authors continue.
This evidence provides a solid foundation for future research exploring new strategies to combat pathogenic microbial biofilms through the potential use of ellagic acid and new natural molecules tailored to the novel protein target, with the aim of mitigating the proliferation of antibiotic-resistant bacteria.
